A prolific nucleator program

چکیده

A prolific nucleator program n studies of Drosophila cells, aurora-A kinase and centrosomin (CNN) are sometimes used as markers for centrosomes, but, other than their localization, relatively little is known about these proteins. On page 757, Terada et al. show that aurora-A and CNN must physically interact to be translocated to the centro-some, and, once there, CNN acts as a powerful microtubule nucleator, placing these two convenient markers at the center of centrosomal function. Using a two-hybrid screen, the authors discovered that aurora-A specifically binds to a COOH-terminal domain of CNN, and RNAi inhibition of either protein prevents the translocation of the other to the centrosome. The NH 2-terminal half of CNN interacts with the ␥-tubulin complex. When overexpressed in fly or mammalian cells, or in an in vitro system, CNN induces microtubule nucleation with striking efficiency. Terada et al. speculate that the two functionally distinct domains of CNN connect the ␥-tubulin complex to aurora-A and the centrosome. Although the natural CNN analogues in mammalian cells have so far eluded detection, the ability of Drosophila CNN to induce microtubule nucleation in mammalian cells underscores the evolutionary conservation of this mechanism. The authors hope to use their new in vitro system to determine how the nucleating site is constructed. ᭿

Actin Nucleation: Spire — Actin Nucleator in a Class of Its Own

The rate limiting step for actin filament polymerisation is nucleation, and two types of nucleator have been described: the Arp2/3 complex and the formins. A recent study has now identified in Spire a third class of actin nucleator. The four short WH2 repeats within Spire bind four consecutive actin monomers to form a novel single strand nucleus for 'barbed end' actin filament elongation.

The Future of a Prolific Variant of Eastern Gamagrass

Eastern gamagrass [Tripsacum dactyloides (L.) L.] is currently being selected for higher forage yield. A prolific variant which can produce up to 20 times the number and 3 times the weight of seeds of a normal plant was found in a wild population and is now instrumental in the breeding program. Could this prolific type spread in wild populations? Vegetative vigor of mature plants was assessed b...

Estimation of the Cultured Cells’ Volume and Surface Area: Application of Stereological Methods on Vero Cells Infected by Rubella Virus

Background: Morphological changes of the cells infected with rubella virus cannot be observed easily. Estimation of the size of the cultured cells can be a valuable parameter in this condition. This study was conducted to find answers to the following questions:How much time after infection with rubella virus, the volume and surface area of the Vero cells and their nuclei get started to change?...
